Editing Parkinson's Disease – KAIST Makes World's First Discovery of an Inflammatory RNA Editing Enzyme

Professor Sonia Gandhi (top right) and Professor Klenerman of the University College London (bottom right) > Parkinson's disease (PD) is a neurodegenerative disorder in which the α-synuclein protein abnormally aggregates within brain cells, causing neuronal damage. Through international collaboration, researchers at KAIST have revealed that RNA editing plays a crucial role in regulating neuroinflammation, a key pathology of Parkinson's disease. KAIST (represented by President Kwang-Hyung Lee) announced on the 27th of April that a research team led by Professor Minee L. Choi from the Department of Brain and Cognitive Sciences, in collaboration with University College London (UCL) and the Francis Crick Institute, discovered that the RNA editing enzyme ADAR1 plays an important role in controlling immune responses in astrocytes, glial cells that trigger protective reactions in the brain, and demonstrated that this mechanism is critically involved in the progression of Parkinson’s disease. Professor Choi's research team created a co-culture model composed of astrocytes and neurons derived from stem cells originating from Parkinson's disease patients, in order to study the inflammatory responses of brain immune cells. They then treated the model with α-synuclein aggregates, which are known to cause Parkinson’s disease, and analyzed how the immune cells' inflammatory responses changed. < Figure 1. Schematic diagram of the inflammatory RNA editing model in Parkinson's disease > As a result, it was found that early pathological forms of α-synuclein, known as oligomers, activated the Toll-like receptor pathway, which acts as a danger sensor in astrocytes, as well as the interferon response pathway, an immune signaling network that combats viruses and pathogens. During this process, the RNA editing enzyme ADAR1 was expressed and transformed into an isoform with an altered protein structure and function. Notably, the RNA editing activity of ADAR1, which normally functions to regulate immune responses during viral infections by converting adenosine (A) to inosine (I) through a process known as A-to-I RNA editing, was found to be abnormally focused on genes that cause inflammation rather than operating under normal conditions. This phenomenon was observed not only in the patient-derived neuron models but also in postmortem brain tissues from actual Parkinson’s disease patients. < Figure 2. Experimental design and inflammatory response induction in astrocytes following treatment with α-synuclein oligomers (abnormally folded protein fragments) > This directly proves that the dysregulation of RNA editing induces chronic inflammatory responses in astrocytes, ultimately leading to neuronal toxicity and pathological progression. This study is significant in that it newly identified the regulation of RNA editing within astrocytes as a key mechanism behind neuroinflammatory responses. In particular, it suggests that ADAR1 could serve as a novel genetic target for the treatment of Parkinson’s disease. It is also noteworthy that the study reflected actual pathological characteristics of patients by utilizing patient-specific induced pluripotent stem cell-based precision models for brain diseases. Professor Minee L. Choi stated, “This study demonstrates that the regulator of inflammation caused by protein aggregation operates at the new layer of RNA editing, offering a completely different therapeutic strategy from existing approaches to Parkinson's disease treatment." She further emphasized, “RNA editing technology could become an important turning point in the development of therapeutics for neuroinflammation.” < Figure 3. KAIST perfectly reproduces Joseon-era Irworobongdo without pigments
Typically, chemical pigments that absorb specific wavelengths of light within the visible spectrum are required to produce colors. However, KAIST researchers have successfully reproduced the Joseon-era Irworobongdo [일월오봉도] painting using ultra-precise color graphics without any chemical pigments, allowing for the permanent and eco-friendly preservation of color graphics without fading or discoloration. < (From left) Chaerim Son, a graduate of the Department of Biochemical Engineering (lead author), Seong Kyeong Nam, a graduate of the PhD program, Jiwoo Lee, a PhD student, and Professor Shin-Hyun Kim > KAIST (represented by President Kwang Hyung Lee) announced on the 26th of February that a research team led by Professor Shinhyun Kim from the Department of Biological and Chemical Engineering had developed a technology that enables high-resolution color graphics without using any chemical pigments by employing hemisphere-shaped microstructures. Morpho butterflies that are brilliant blue in color or Panther chameleons that change skin color exhibit coloration without chemical pigments, as ordered nanostructures within a material reflect visible light through optical interference. Since structural colors arise from physical structures rather than chemical substances, a single material can produce a wide range of colors. However, the artificial implementation of structural coloration is highly challenging due to the complexity of creating ordered nanostructures. Additionally, it is difficult to produce a variety of colors and to pattern them precisely into complex designs. < Figure 1. Principle of structural color expression using micro-hemispheres (left) and method of forming micro-hemisphere patterns based on photolithography (right) > Professor Kim’s team overcame these challenges by using smooth-surfaced hemispherical microstructures instead of ordered nanostructures, enabling the high-precision patterning of diverse structural colors. When light enters the inverted hemispherical microstructures, the portion of light entering from the sides undergoes total internal reflection along the curved surface, creating retroreflection. When the hemisphere diameter is approximately 10 micrometers (about one-tenth the thickness of a human hair), light traveling along different reflection paths interferes within the visible spectrum, producing structural coloration. < Figure 2. “Irworobongdo”, the Painting of the Sun, Moon, and the Five Peaks, reproduced in fingernail size without pigment using approximately 200,000 micro-hemispheres > The structural color can be tuned by adjusting the size of the hemispheres. By arranging hemispheres of varying sizes, much like mixing paints on a palette, an infinite range of colors can be generated. To precisely pattern microscale hemispheres of different sizes, the research team employed photolithography* using positive photoresists** commonly used in semiconductor processing. They first patterned photoresists into micropillar structures, then induced reflow*** by heating the material, forming hemispherical microstructures. *Photolithography: A technique used in semiconductor fabrication to pattern microscale structures. **Positive photoresist: A photosensitive polymer that dissolves more easily in a developer solution after exposure to ultraviolet light. ***Reflow: A process in which a polymer material softens and reshapes into a curved structure when heated. This method enables the formation of hemisphere-shaped microstructures with the desired sizes and colors in a single-step fabrication process. It also allows for the reproduction of arbitrary color graphics using a single material without any pigments. The ultra-precise color graphics created with this technique can exhibit color variations depending on the angle of incident light or the viewing perspective. The pattern appears colored from one direction while remaining transparent from the opposite side, exhibiting a Janus effect. These structural color graphics achieve resolution comparable to cutting-edge LED displays, allowing complex color images to be captured within a fingernail-sized area and projected onto large screens. < Figure 3. “Irworobongdo” that displays different shades depending on the angle of light and viewing direction > Professor Shinhyun Kim, who led the research, stated, “Our newly developed pigment-free color graphics technology can serve as an innovative method for artistic expression, merging art with advanced materials. Additionally, it holds broad application potential in optical devices and sensors, anti-counterfeiting materials, aesthetic photocard printing, and many other fields.” This research, with KAIST researcher Chaerim Son as the first author, was published in the prestigious materials science journal Advanced Materials on February 5. KAIST Professor Uichin Lee Receives Distinguished Paper Award from ACM

Professor Uichin Lee (left) receiving the award > KAIST (President Kwang Hyung Lee) announced on the 25th of October that Professor Uichin Lee’s research team from the School of Computing received the Distinguished Paper Award at the International Joint Conference on Pervasive and Ubiquitous Computing and International Symposium on Wearable Computing (Ubicomp / ISWC) hosted by the Association for Computing Machinery (ACM) in Melbourne, Australia on October 8. The ACM Ubiquitous Computing Conference is the most prestigious international conference where leading universities and global companies from around the world present the latest research results on ubiquitous computing and wearable technologies in the field of human-computer interaction (HCI). The main conference program is composed of invited papers published in the Proceedings of the ACM (PACM) on Interactive, Mobile, Wearable and Ubiquitous Technologies (IMWUT), which covers the latest research in the field of ubiquitous and wearable computing. The Distinguished Paper Award Selection Committee selected eight papers among 205 papers published in Vol. 7 of the ACM Proceedings (PACM IMWUT) that made outstanding and exemplary contributions to the research community. The committee consists of 16 prominent experts who are current and former members of the journal's editorial board which made the selection after a rigorous review of all papers for a period that stretched over a month. < Figure 1. BeActive mobile app to promote physical activity to form active lifestyle habits > The research that won the Distinguished Paper Award was conducted by Dr. Junyoung Park, a graduate of the KAIST Graduate School of Data Science, as the 1st author, and was titled “Understanding Disengagement in Just-in-Time Mobile Health Interventions” Professor Uichin Lee’s research team explored user engagement of ‘Just-in-Time Mobile Health Interventions’ that actively provide interventions in opportune situations by utilizing sensor data collected from health management apps, based on the premise that these apps are aptly in use to ensure effectiveness. < Figure 2. Traditional user-requested digital behavior change intervention (DBCI) delivery (Pull) vs. Automatic transmission (Push) for Just-in-Time (JIT) mobile DBCI using smartphone sensing technologies > The research team conducted a systematic analysis of user disengagement or the decline in user engagement in digital behavior change interventions. They developed the BeActive system, an app that promotes physical activities designed to help forming active lifestyle habits, and systematically analyzed the effects of users’ self-control ability and boredom-proneness on compliance with behavioral interventions over time. The results of an 8-week field trial revealed that even if just-in-time interventions are provided according to the user’s situation, it is impossible to avoid a decline in participation. However, for users with high self-control and low boredom tendency, the compliance with just-in-time interventions delivered through the app was significantly higher than that of users in other groups. In particular, users with high boredom proneness easily got tired of the repeated push interventions, and their compliance with the app decreased more quickly than in other groups. < Figure 3. Just-in-time Mobile Health Intervention: a demonstrative case of the BeActive system: When a user is identified to be sitting for more than 50 mins, an automatic push notification is sent to recommend a short active break to complete for reward points. > Professor Uichin Lee explained, “As the first study on user engagement in digital therapeutics and wellness services utilizing mobile just-in-time health interventions, this research provides a foundation for exploring ways to empower user engagement.” He further added, “By leveraging large language models (LLMs) and comprehensive context-aware technologies, it will be possible to develop user-centered AI technologies that can significantly boost engagement." < Figure 4. Engagement in digital health apps consists of (1) engagement in using digital health apps and (2) engagement in behavioral interventions provided by digital health apps, i.e., compliance with behavioral interventions. KAIST Develops a Fire-risk Free Self-Powered Hydrogen Production System
KAIST researchers have developed a new hydrogen production system that overcomes the current limitations of green hydrogen production. By using a water-splitting system with an aqueous electrolyte, this system is expected to block fire risks and enable stable hydrogen production. KAIST (represented by President Kwang Hyung Lee) announced on the 22nd of October that a research team led by Professor Jeung Ku Kang from the Department of Materials Science and Engineering developed a self-powered hydrogen production system based on a high-performance zinc-air battery*. *Zinc-air battery: A primary battery that absorbs oxygen from the air and uses it as an oxidant. Its advantage is long life, but its low electromotive force is a disadvantage. Hydrogen (H₂) is a key raw material for synthesizing high-value-added substances, and it is gaining attention as a clean fuel with an energy density (142 MJ/kg) more than three times higher than traditional fossil fuels (gasoline, diesel, etc.). However, most current hydrogen production methods impose environmental burden as they emit carbon dioxide (CO₂). While green hydrogen can be produced by splitting water using renewable energy sources such as solar cells and wind power, these sources are subject to irregular power generation due to weather and temperature fluctuations, leading to low water-splitting efficiency. To overcome this, air batteries that can emit sufficient voltage (greater than 1.23V) for water splitting have been gaining attention. However, achieving sufficient capacity requires expensive precious metal catalysts and the performance of the catalyst materials becomes significantly degraded during prolonged charge and discharge cycles. Thus, it is essential to develop catalysts that are effective for the water-splitting reactions (oxygen and hydrogen evolution) and materials that can stabilize the repeated charge and discharge reactions (oxygen reduction and evolution) in zinc-air battery electrodes. In response, Professor Kang's research team proposed a method to synthesize a non-precious metal catalyst material (G-SHELL) that is effective for three different catalytic reactions (oxygen evolution, hydrogen evolution, and oxygen reduction) by growing nano-sized, metal-organic frameworks on graphene oxide. The team incorporated the developed catalyst material into the air cathode of a zinc-air battery, confirming that it achieved approximately five times higher energy density (797Wh/kg), high power characteristics (275.8mW/cm²), and long-term stability even under repeated charge and discharge conditions compared to conventional batteries. Additionally, the zinc-air battery, which operates using an aqueous electrolyte, is safe from fire risks. It is expected that this system can be applied as a next-generation energy storage device when linked with water electrolysis systems, offering an environmentally friendly method for hydrogen production. < Figure 1. A KAIST-SNUH Team Devises a Way to Make Mathematical Predictions to find Metabolites Related to Somatic Mutations in Cancers
Cancer is characterized by abnormal metabolic processes different from those of normal cells. Therefore, cancer metabolism has been extensively studied to develop effective diagnosis and treatment strategies. Notable achievements of cancer metabolism studies include the discovery of oncometabolites* and the approval of anticancer drugs by the U.S. Food and Drug Administration (FDA) that target enzymes associated with oncometabolites. Approved anticancer drugs such as ‘Tibsovo (active ingredient: ivosidenib)’ and ‘Idhifa (active ingredient: enasidenib)’ are both used for the treatment of acute myeloid leukemia. Despite such achievements, studying cancer metabolism, especially oncometabolites, remains challenging due to time-consuming and expensive methodologies such as metabolomics. Thus, the number of confirmed oncometabolites is very small although a relatively large number of cancer-associated gene mutations have been well studied. *Oncometabolite: A metabolite that shows pro-oncogenic function when abnormally accumulated in cancer cells. An oncometabolite is often generated as a result of gene mutations, and this accumulation promotes the growth and survival of cancer cells. Representative oncometabolites include 2-hydroxyglutarate, succinate, and fumarate. On March 18th, a KAIST research team led by Professor Hyun Uk Kim from the Department of Chemical and Biomolecular Engineering developed a computational workflow that systematically predicts metabolites and metabolic pathways associated with somatic mutations in cancer through collaboration with research teams under Prof Youngil Koh, Prof. Hongseok Yun, and Prof. Chang Wook Jeong from Seoul National University Hospital. The research teams have successfully reconstructed patient-specific genome-scale metabolic models (GEMs)* for 1,043 cancer patients across 24 cancer types by integrating publicly available cancer patients’ transcriptome data (i.e., from international cancer genome consortiums such as PCAWG and TCGA) into a generic human GEM. The resulting patient-specific GEMs make it possible to predict each patient’s metabolic phenotypes. *Genome-scale metabolic model (GEM): A computational model that mathematically describes all of the biochemical reactions that take place inside a cell. It allows for the prediction of the cell’s metabolic phenotypes under various genetic and/or environmental conditions. < Figure 1. Schematic diagram of a computational methodology for predicting metabolites and metabolic pathways associated with cancer somatic mutations. of a computational methodology for predicting metabolites and metabolic pathways associated with cancer somatic mutations. > The team developed a four-step computational workflow using the patient-specific GEMs from 1,043 cancer patients and somatic mutation data obtained from the corresponding cancer patients. This workflow begins with the calculation of the flux-sum value of each metabolite by simulating the patient-specific GEMs. The flux-sum value quantifies the intracellular importance of a metabolite. Next, the workflow identifies metabolites that appear to be significantly associated with specific gene mutations through a statistical analysis of the predicted flux-sum data and the mutation data. Finally, the workflow selects altered metabolic pathways that significantly contribute to the biosynthesis of the predicted oncometabolite candidates, ultimately generating metabolite-gene-pathway sets as an output. The two co-first authors, Dr. GaRyoung Lee (currently a postdoctoral fellow at the Dana-Farber Cancer Institute and Harvard Medical School) and Dr. Sang Mi Lee (currently a postdoctoral fellow at Harvard Medical School) said, “The computational workflow developed can systematically predict how genetic mutations affect cellular metabolism through metabolic pathways. Importantly, it can easily be applied to different types of cancer based on the mutation and transcriptome data of cancer patient cohorts.” Prof. Kim said, “The computational workflow and its resulting prediction outcomes will serve as the groundwork for identifying novel oncometabolites and for facilitating the development of various treatment and diagnosis strategies”. This study, which was supported by the National Research Foundation of Korea, has been published online in Genome Biology, a representative journal in the field of biotechnology and genetics, under the title "Prediction of metabolites associated with somatic mutations in cancers by using genome‑scale metabolic models and mutation data".

Team KAIST placed among top two at MBZIRC Maritime Grand Challenge
Representing Korean Robotics at Sea: KAIST’s 26-month strife rewarded Team KAIST placed among top two at MBZIRC Maritime Grand Challenge - Team KAIST, composed of students from the labs of Professor Jinwhan Kim of the Department of Mechanical Engineering and Professor Hyunchul Shim of the School of Electrical and Engineering, came through the challenge as the first runner-up winning the prize money totaling up to $650,000 (KRW 860 million). - Successfully led the autonomous collaboration of unmanned aerial and maritime vehicles using cutting-edge robotics and AI technology through to the final round of the competition held in Abu Dhabi from January 10 to February 6, 2024. KAIST (President Kwang-Hyung Lee), reported on the 8th that Team KAIST, led by students from the labs of Professor Jinwhan Kim of the Department of Mechanical Engineering and Professor Hyunchul Shim of the School of Electrical Engineering, with Pablo Aviation as a partner, won a total prize money of $650,000 (KRW 860 million) at the Maritime Grand Challenge by the Mohamed Bin Zayed International Robotics Challenge (MBZIRC), finishing first runner-up. This competition, which is the largest ever robotics competition held over water, is sponsored by the government of the United Arab Emirates and organized by ASPIRE, an organization under the Abu Dhabi Ministry of Science, with a total prize money of $3 million. In the competition, which started at the end of 2021, 52 teams from around the world participated and five teams were selected to go on to the finals in February 2023 after going through the first and second stages of screening. The final round was held from January 10 to February 6, 2024, using actual unmanned ships and drones in a secluded sea area of 10 km2 off the coast of Abu Dhabi, the capital of the United Arab Emirates. A total of 18 KAIST students and Professor Jinwhan Kim and Professor Hyunchul Shim took part in this competition at the location at Abu Dhabi. Team KAIST will receive $500,000 in prize money for taking second place in the final, and the team’s prize money totals up to $650,000 including $150,000 that was as special midterm award for finalists. The final mission scenario is to find the target vessel on the run carrying illegal cargoes among many ships moving within the GPS-disabled marine surface, and inspect the deck for two different types of stolen cargo to recover them using the aerial vehicle to bring the small cargo and the robot manipulator topped on an unmanned ship to retrieve the larger one. The true aim of the mission is to complete it through autonomous collaboration of the unmanned ship and the aerial vehicle without human intervention throughout the entire mission process. In particular, since GPS cannot be used in this competition due to regulations, Professor Jinwhan Kim's research team developed autonomous operation techniques for unmanned ships, including searching and navigating methods using maritime radar, and Professor Hyunchul Shim's research team developed video-based navigation and a technology to combine a small autonomous robot with a drone. The final mission is to retrieve cargo on board a ship fleeing at sea through autonomous collaboration between unmanned ships and unmanned aerial vehicles without human intervention. The overall mission consists the first stage of conducting the inspection to find the target ship among several ships moving at sea and the second stage of conducting the intervention mission to retrieve the cargoes on the deck of the ship. Each team was given a total of three opportunities, and the team that completed the highest-level mission in the shortest time during the three attempts received the highest score. In the first attempt, KAIST was the only team to succeed in the first stage search mission, but the competition began in earnest as the Croatian team also completed the first stage mission in the second attempt. As the competition schedule was delayed due to strong winds and high waves that continued for several days, the organizers decided to hold the finals with the three teams, including the Team KAIST and the team from Croatia’s the University of Zagreb, which completed the first stage of the mission, and Team Fly-Eagle, a team of researcher from China and UAE that partially completed the first stage. The three teams were given the chance to proceed to the finals and try for the third attempt, and in the final competition, the Croatian team won, KAIST took the second place, and the combined team of UAE-China combined team took the third place. The final prize to be given for the winning team is set at $2 million with $500,000 for the runner-up team, and $250,000 for the third-place. Professor Jinwhan Kim of the Department of Mechanical Engineering, who served as the advisor for Team KAIST, said, “I would like to express my gratitude and congratulations to the students who put in a huge academic and physical efforts in preparing for the competition over the past two years. NYU-KAIST Global AI & Digital Governance Conference Held

Mills, and KAIST President Kwang Hyung Lee, KAIST co-hosted the NYU-KAIST Global AI & Digital Governance Conference at the Paulson Center of New York University (NYU) in New York City, USA on September 21st, 9:30 pm. At the conference, KAIST and NYU discussed the direction and policies for ‘global AI and digital governance’ with participants of upto 300 people which includes scholars, professors, and students involved in the academic field of AI and digitalization from both Korea and the United States and other international backgrounds. This conference was a forum of an international discussion that sought new directions for AI and digital technology take in the future and gathered consensus on regulations. Following a welcoming address by KAIST President, Kwang Hyung Lee and a congratulatory message from the Minister of Science and ICT, Jong-ho Lee, a panel discussion was held, moderated by Professor Matthew Liao, a graduate of Princeton and Oxford University, currently serving as a professor at NYU and the director at the Center for Bioethics of the NYU School of Global Public Health. Six prominent scholars took part in the panel discussion. Prof. Kyung-hyun Cho of NYU Applied Mathematics and Data Science Center, a KAIST graduate who has joined the ranks of the world-class in AI language models and Professor Jong Chul Ye, the Director of Promotion Council for Digital Health at KAIST, who is leading innovative research in the field of medical AI working in collaboration with major hospitals at home and abroad was on the panel. Additionally, Professor Luciano Floridi, a founding member of the Yale University Center for Digital Ethics, Professor Shannon Vallor, the Baillie Gifford Professor in the Ethics of Data and Artificial Intelligence at the University of Edinburgh of the UK, Professor Stefaan Verhulst, a Co-Founder and the DIrector of GovLab‘s Data Program at NYU’s Tandon School of Engineering, and Professor Urs Gasser, who is in charge of public policy, governance and innovative technology at the Technical University of Munich, also participated. Professor Matthew Liao from NYU led the discussion on various topics such as the ways to to regulate AI and digital technologies; the concerns about how deep learning technology being developed in medicinal purposes could be used in warfare; the scope of responsibilities Al scientists' responsibility should carry in ensuring the usage of AI are limited to benign purposes only; the effects of external regulation on the AI model developers and the research they pursue; and on the lessons that can be learned from the regulations in other fields. During the panel discussion, there was an exchange of ideas about a system of standards that could harmonize digital development and regulatory and social ethics in today’s situation in which digital transformation accelerates technological development at a global level, there is a looming concern that while such advancements are bringing economic vitality it may create digital divides and probles like manipulation of public opinion. Professor Jong-cheol Ye of KAIST (Director of the Promotion Council for Digital Health), in particular, emphasized that it is important to find a point of balance that does not hinder the advancements rather than opting to enforcing strict regulations. < Photo 2. Earlier, KAIST signed a cooperation agreement with NYU to build a joint campus, June last year and held a plaque presentation ceremony for the KAIST NYU Joint Campus last September to promote joint research between the two universities. KAIST is currently conducting joint research with NYU in nine fields, including AI and digital research. The KAIST-NYU Joint Campus was conceived with the goal of building an innovative sandbox campus centering aroung science, technology, engineering, and mathematics (STEM) combining NYU's excellent humanities and arts as well as basic science and convergence research capabilities with KAIST's science and technology. KAIST has contributed to the development of Korea's industry and economy through technological innovation aiding in the nation’s transformation into an innovative nation with scientific and technological prowess. KAIST holds its first ‘KAIST Tech Fair’ in New York, USA

The strength of their solution is that logistics robots equipped with AI reinforced learning technology can respond to processes and environmental changes on their own, minimizing maintenance costs and the system can achieve excellent performance even with a small amount of data when it is combined with the digital twin technology the company has developed on its own. A student startup, ‘Aniai’, is entering the US market, the home of hamburgers, with hamburger patty automation equipments and solutions. This is a robot kitchen startup founded by its CEO Gunpil Hwang, a graduate of KAIST’s School of Electrical Engineering which gathered together the experts in the fields of robot control, design, and artificial intelligence and cognitive technology to develop technology to automatically cook hamburger patties. At the touch of a button, both sides of the patty are cooked simultaneously for consistent taste and quality according to the set condition. Since it can cook about 200 dishes in an hour, it is attracting attention as a technology that can not only solve manpower shortages but also accelerate the digital transformation of the restaurant industry. KAIST debuts “DreamWaQer” - a quadrupedal robot that can walk in the dark
- The team led by Professor Hyun Myung of the School of Electrical Engineering developed “DreamWaQ”, a deep reinforcement learning-based walking robot control technology that can walk in an atypical environment without visual and/or tactile information - Utilization of “DreamWaQ” technology can enable mass production of various types of “DreamWaQers” - Expected to be used in exploration of atypical environment involving unique circumstances such as disasters by fire. A team of Korean engineering researchers has developed a quadrupedal robot technology that can climb up and down the steps and moves without falling over in uneven environments such as tree roots without the help of visual or tactile sensors even in disastrous situations in which visual confirmation is impeded due to darkness or thick smoke from the flames. KAIST (President Kwang Hyung Lee) announced on the 29th of March that Professor Hyun Myung's research team at the Urban Robotics Lab in the School of Electrical Engineering developed a walking robot control technology that enables robust 'blind locomotion' in various atypical environments. < (From left) Prof. Hyun Myung, Doctoral Candidates I Made Aswin Nahrendra, Byeongho Yu, and Minho Oh. In the foreground is the DreamWaQer, a quadrupedal robot equipped with DreamWaQ technology. > The KAIST research team developed "DreamWaQ" technology, which was named so as it enables walking robots to move about even in the dark, just as a person can walk without visual help fresh out of bed and going to the bathroom in the dark. With this technology installed atop any legged robots, it will be possible to create various types of "DreamWaQers". Existing walking robot controllers are based on kinematics and/or dynamics models. This is expressed as a model-based control method. In particular, on atypical environments like the open, uneven fields, it is necessary to obtain the feature information of the terrain more quickly in order to maintain stability as it walks. However, it has been shown to depend heavily on the cognitive ability to survey the surrounding environment. In contrast, the controller developed by Professor Hyun Myung's research team based on deep reinforcement learning (RL) methods can quickly calculate appropriate control commands for each motor of the walking robot through data of various environments obtained from the simulator. Whereas the existing controllers that learned from simulations required a separate re-orchestration to make it work with an actual robot, this controller developed by the research team is expected to be easily applied to various walking robots because it does not require an additional tuning process. DreamWaQ, the controller developed by the research team, is largely composed of a context estimation network that estimates the ground and robot information and a policy network that computes control commands. The context-aided estimator network estimates the ground information implicitly and the robot’s status explicitly through inertial information and joint information. This information is fed into the policy network to be used to generate optimal control commands. Both networks are learned together in the simulation. While the context-aided estimator network is learned through supervised learning, the policy network is learned through an actor-critic architecture, a deep RL methodology. The actor network can only implicitly infer surrounding terrain information. In the simulation, the surrounding terrain information is known, and the critic, or the value network, that has the exact terrain information evaluates the policy of the actor network. This whole learning process takes only about an hour in a GPU-enabled PC, and the actual robot is equipped with only the network of learned actors. Without looking at the surrounding terrain, it goes through the process of imagining which environment is similar to one of the various environments learned in the simulation using only the inertial sensor (IMU) inside the robot and the measurement of joint angles. If it suddenly encounters an offset, such as a staircase, it will not know until its foot touches the step, but it will quickly draw up terrain information the moment its foot touches the surface. Then the control command suitable for the estimated terrain information is transmitted to each motor, enabling rapidly adapted walking. The DreamWaQer robot walked not only in the laboratory environment, but also in an outdoor environment around the campus with many curbs and speed bumps, and over a field with many tree roots and gravel, demonstrating its abilities by overcoming a staircase with a difference of a height that is two-thirds of its body. In addition, regardless of the environment, the research team confirmed that it was capable of stable walking ranging from a slow speed of 0.3 m/s to a rather fast speed of 1.0 m/s. The results of this study were produced by a student in doctorate course, I Made Aswin Nahrendra, as the first author, and his colleague Byeongho Yu as a co-author. Synthetic sRNAs to knockdown genes in medical and industrial bacteria
Bacteria are intimately involved in our daily lives. These microorganisms have been used in human history for food such as cheese, yogurt, and wine, In more recent years, through metabolic engineering, microorganisms been used extensively as microbial cell factories to manufacture plastics, feed for livestock, dietary supplements, and drugs. However, in addition to these bacteria that are beneficial to human lives, pathogens such as Pneumonia, Salmonella, and Staphylococcus that cause various infectious diseases are also ubiquitously present. It is important to be able to metabolically control these beneficial industrial bacteria for high value-added chemicals production and to manipulate harmful pathogens to suppress its pathogenic traits. KAIST (President Kwang Hyung Lee) announced on the 10th that a research team led by Distinguished Professor Sang Yup Lee of the Department of Biochemical Engineering has developed a new sRNA tool that can effectively inhibit target genes in various bacteria, including both Gram-negative and Gram-positive bacteria. To address this issue, a research team led by Distinguished Professor Lee Sang Yup Lee of the Department of Chemical and Biomolecular Engineering at KAIST developed a new sRNA platform that can effectively suppress target genes in various bacteria, including both Gram-negative and positive bacteria. The research team surveyed thousands of microbial-derived sRNA systems in the microbial database, and eventually designated the sRNA system derived from 'Bacillus subtilis' that showed the highest gene knockdown efficiency, and designated it as “Broad-Host-Range sRNA”, or BHR-sRNA. A similar well-known system is the CRISPR interference (CRISPRi) system, which is a modified CRISPR system that knocks down gene expression by suppressing the gene transcription process. However, the Cas9 protein in the CRISPRi system has a very high molecular weight, and there have been reports growth inhibition in bacteria. The BHR-sRNA system developed in this study did not affect bacterial growth while showing similar gene knockdown efficiencies to CRISPRi. < Figure 1. a) Schematic illustration demonstrating the mechanism of syntetic sRNA b) Phylogenetic tree of the 16 Gram-negative and Gram-positive bacterial species tested for gene knockdown by the BHR-sRNA system. > To validate the versatility of the BHR-sRNA system, 16 different gram-negative and gram-positive bacteria were selected and tested, where the BHR-sRNA system worked successfully in 15 of them. In addition, it was demonstrated that the gene knockdown capability was more effective than that of the existing E. coli-based sRNA system in 10 bacteria. The BHR-sRNA system proved to be a universal tool capable of effectively inhibiting gene expression in various bacteria. In order to address the problem of antibiotic-resistant pathogens that have recently become more serious, the BHR-sRNA was demonstrated to suppress the pathogenicity by suppressing the gene producing the virulence factor. By using BHR-sRNA, biofilm formation, one of the factors resulting in antibiotic resistance, was inhibited by 73% in Staphylococcus epidermidis a pathogen that can cause hospital-acquired infections. Antibiotic resistance was also weakened by 58% in the pneumonia causing bacteria Klebsiella pneumoniae. In addition, BHR-sRNA was applied to industrial bacteria to develop microbial cell factories to produce high value-added chemicals with better production performance. Notably, superior industrial strains were constructed with the aid of BHR-sRNA to produce the following chemicals: valerolactam, a raw material for polyamide polymers, methyl-anthranilate, a grape-flavor food additive, and indigoidine, a blue-toned natural dye. The BHR-sRNA developed through this study will help expedite the commercialization of bioprocesses to produce high value-added compounds and materials such as artificial meat, jet fuel, health supplements, pharmaceuticals, and plastics. It is also anticipated that to help eradicating antibiotic-resistant pathogens in preparation for another upcoming pandemic. “In the past, we could only develop new tools for gene knockdown for each bacterium, but now we have developed a tool that works for a variety of bacteria” said Distinguished Professor Sang Yup Lee. KAIST gearing up to train physician-scientists and BT Professionals joining hands with Boston-based organizations
KAIST (President Kwang Hyung Lee) announced on the 29th that it has signed MOUs with Massachusetts General Hospital, a founding member of the Mass General Brigham health care system and a world-class research-oriented hospital, and Moderna, a biotechnology company that developed a COVID-19 vaccine at the Langham Hotel in Boston, MA, USA on the morning of April 28th (local time). The signing ceremony was attended by officials from each institution joined by others headed by Minister LEE Young of the Korean Ministry of SMEs and Startups (MSS), and Commissioner LEE Insil of the Korean Intellectual Property Office. < Photo 1. Photo from the Signing of MOU between KAIST-Harvard University Massachusetts General Hospital and KAIST-Moderna > Mass General is the first and largest teaching hospital of Harvard Medical School in Boston, USA, and it is one of the most innovative hospitals in the world being the alma mater of more than 13 Nobel Prize winners and the home of the Mass General Research Institute, the world’s largest hospital-based research program that utilizes an annual research budget of more than $1.3 billion. KAIST signed a general agreement to explore research and academic exchange with Mass General in September of last year and this MOU is a part of its follow-ups. Mass General works with Harvard and the Massachusetts Institute of Technology (MIT), as well as local hospitals, to support students learn the theories of medicine and engineering, and gain rich clinical research experience. Through this MOU, KAIST will explore cooperation with an innovative ecosystem created through the convergence of medicine and engineering. In particular, KAIST’s goal is to develop a Korean-style training program and implement a differentiated educational program when establishing the science and technology-oriented medical school in the future by further strengthening the science and engineering part of the training including a curriculum on artificial intelligence (AI) and the likes there of. Also, in order to foster innovative physician-scientists, KAIST plans to pursue cooperation to develop programs for exchange of academic and human resources including programs for student and research exchanges and a program for students of the science and technology-oriented medical school at KAIST to have a chance to take part in practical training at Mass General. David F.M. Brown, MD, Mass General President, said, “The collaboration with KAIST has a wide range of potentials, including advice on training of physician-scientists, academic and human resource exchanges, and vitalization of joint research by faculty from both institutions. Through this agreement, we will be able to actively contribute to global cooperation and achieve mutual goals.” Meanwhile, an MOU between KAIST and Moderna was also held on the same day. Its main focus is to foster medical experts in cooperation with KAIST Graduate School of Medical Science and Engineering (GSMSE), and plans to cooperate in various ways in the future, including collaborating for development of vaccine and new drugs, virus research, joint mRNA research, and facilitation of technology commercialization. In over 10 years since its inception, Moderna has transformed from a research-stage company advancing programs in the field of messenger RNA (mRNA) to an enterprise with a diverse clinical portfolio of vaccines and therapeutics across seven modalities. UAE Space Program Leaders named to be the 1st of the honorees of KAIST Alumni Association's special recognition for graduates of foreign nationality
The KAIST Alumni Association (Chairman, Chil-Hee Chung) announced on the 12th that the winners of the 2023 KAIST Distinguished Alumni Award and International Alumni Award has been selected. The KAIST Distinguished Alumni Award, which produced the first recipient in 1992, is an award given to alumni who have contributed to the development of the nation and society, or who have glorified the honor of their alma mater with outstanding academic achievements and social and/or communal contributions. On a special note, this year, there has been an addition to the honors, “the KAIST Distinguished International Alumni Award” to honor and encourage overseas alumni who are making their marks in the international community that will boost positive recognition of KAIST in the global setting and will later become a bridge that will expedite Korea's international efforts in the future. As of 2022, the number of international students who succeeded in earning KAIST degrees has exceeded 1,700, and they are actively doing their part back in their home countries as leaders in various fields in which they belong, spanning from science and technology, to politics, industry and other corners of the society. Currently, the three alums are in directorship of the Mohammed Bin Rashid Space Centre (MBRSC) with Mr. Omran Sharaf, who has recently been appointed as the Assistant Minister in charge of Advanced Science and Technology at the UAE Ministry of Foreign Affairs and International Cooperation, being the Project Director of the Emirates Mars Mission of MBRSC and Mr. Amer Al Sayegh in the Director General position in charge of Space Project and Mr. Mohammed Al Harmi, the Director General of Administration, at MBRSC. They received technology transfer from “SatRec I”, Korea's first satellite system exporter and KAIST alumni company, for about 10 years from 2006, while carrying out their master’s studies at the same time. Afterwards, they returned to UAE to lead the Emirates Mars Mission, which is already showing tangible progress including the successful launch of the Mars probe "Amal" (ال امل, meaning ‘Hope’ in Arabic), which was the first in the Arab world and the fifth in the world to successfully enter into orbit around Mars, and the UAE’s first independently developed Earth observation satellite "KhalifaSat". An official from the KAIST Alumni Association said, "We selected the Distinguished International Alumni after evaluating their industrious leadership in promoting various space industry strategies, ranging from the development of Mars probes and Earth observation satellites, as well as lunar exploration, asteroid exploration, and Mars residence plans." Ltd., Jong Seung Park, the President of Agency for Defense Development (ADD), and Kyunghyun Cho, a Professor of New York University (NYU), were selected as the winners of the “Distinguished Alumni Award”. Mr. Joo-Sun Choi (Electrical and Electronic Engineering, M.S. in 1989, Ph.D. in 1995), the CEO of Samsung Display, led the successful development and mass-production of the world's first ultra-high-definition QD-OLED Displays, and preemptively transformed the structure of business of the industry and has been leading the way in technological innovation. Mr. Jung Goo Cho (Electrical and Electronic Engineering, M.S. in 1988, Ph.D. in 1992), the CEO of Green Power Co. Ltd., developed wireless power technology for the first time in Korea in the early 2000s and applied it to semiconductor/display lines and led the wireless power charging technology in various fields, such as developing KAIST On-Line Electric Vehicles (OLEV) and commercializing the world's first wireless charger for 11kW electric vehicles. Mr. Jong Seung Park (Mechanical Engineering, M.S. in 1988, Ph.D., in 1991), The President of ADD is an expert with abundant science and technology knowledge and organizational management capabilities. He is contributing greatly to national defense and security through science and technology. Mr. Kyunghyun Cho (Computer Science, B.S., in 2009), the Professor of Computer Science and Data Science at NYU, is a world-renowned expert in Artificial Intelligence (AI), advancing the concept of 'Neural Machine Translation' in the field of natural language processing, to make great contributions to AI translation technology and related industries. Chairman Chil-Hee Chung, the 26th Chair of KAIST Alumni Association “As each year goes by, I feel that the influence of KAIST alumni goes beyond science and technology to affect our society as a whole.” He went on to say, “This year, as it was more meaningful to extend the award to honor the international members of our Alums, we look forward to seeing more of our alumni continuing their social and academic endeavors to play an active role in the global stage in taking on the global challenges.” The Ceremony for KAIST Distinguished Alumni and International Alumni Award Honorees will be conducted at the Annual New Year’s Event of KAIST Alumni Association for 2023 to be held on Friday, January 13th, at the Grand InterContinental Seoul Parnas.
